Secretion of Prolactin Following Three Dimensional Culture of Human Endometrial Tissue.
[摘要] Introduction::Endometriosisisthepresenceofendometrialglandularandstromalcellsoutsideoftheuterinecavity.Thisdiseaseisfoundinabout10%ofwomenofthereproductiveageandinupto50%ofwomenwithinfertility.Surgerycontinuesasthefirst-linetreatmenttoeradicateendometrioticlesionsbutrecurrenceoftheconditionoccursinupto47%ofwomen.MaterialsandMethods::Theaimofthepresentstudyistodetermineprolactinlevelsinhumancultureofendometrialfragment.EndometrialbiopsyinPremenopausalpatient’swomenreferredtoTorontoCenterforAdvancedReproductiveTechnology(T.C.A.R.T)forinfertilitytreatmentsuchasuterinemyomasorovariancyst.Endometrialsampleswerecollectedfromatotaloftennormalovulatingwomenoncycledays19-24.Thebiopsieswereobtainedfromthefundalregionoftheuterinecavity.Tentissuefragmentswereculturedbythreedimensionalmethodsforeachpatient.Supernatantfluidsamplewascollectedfromendometrialsampleswhichwereculturedinathree-dimensionalfibrinmatrix.Results::LevelofprolactininSupernatantfluidofendometrialsampleswereplacedinathree-dimensionalfibrinmatrixculturesystemweredetermined.ThesedatashowedCellproliferationwasobservedin91%ofthewells.Angiogenesiswasobservedin51wellsthatshowedcellproliferation(56%).ThelevelofProlactininthesupernatantfluidofwellsthatshowedangiogenesiswereincreased(P#60;0.05)comparetosupernatantfluidofwellsthatdidn’tshowangiogenesis.Conclusion::Prolactinmightplayanimportantroleinpromotingneovascularizationandcellproliferationinestablishmentendometriosis.Prolactinisinvolvedintheregenerationoftheendometriumandthegrowthofendometriosis.
