Typing of Aspergillus Fumigatus and Aspergillus Niger Strains by Random Amplification of Polymorphic DNA Analysis Using a Six Primer Set.
[摘要] Abstract::DifferentiatingandtypingclinicalandenvironmentalisolatesofAspergillusstrainsarenecessaryforepidemiologicalstudiesandcouldcontributetothesolutionofseveralpertinentclinicalproblems.WeinvestigatedtheutilityoftheRAPD-PCRtechniquefortypingtwenty-sixstrainsofAspergillusfumigatusandtenadditionalclinicalandenvironmentalAspergillusfumigatusandAspergillusnigerisolates.FungalgenomicDNAwasextractedusingglassbeaddisruptionandRAPDanalysiswasperformedbyusingasixindividual(R108,R151,UBC90)andcombined(R108+R151,R108+UBS90,R151+UBC90)primerset.PrimerpairR108+UBC90demonstratedthehighestdiscriminatorypowerwithcollectionstrains,whereasprimerR151displayedthehighestdegreeofdiscriminationpowerwithclinicalandenvironmentalisolates.Bothprimersetsdetectedseventypesofstrains.ItwasfoundthatprimerR151,onitsownandincombinationwithprimerUBC90,sharedarelativerelationshipbetweenclinicalandenvironmentalA.fumigatusisolates.WefoundthatalthoughRAPD-PCRanalysiscanbeappliedasasimple,rapid,andveryeffectivemethodfordifferentiatingAspergillusstrains,selectingthebestprimersisacriticalsteptoreachthedesirablediscriminatorypower.
