Evaluation of Enzyme Linked Immunosorbent Assay (ELISA) and Dot ELISA for Diagnosis of Amoebiasis.
[摘要] Introduction:Thisstudywasconductedtoevaluatetheusefulnessofdotenzyme-linkedimmunosorbentassay(dotELISA)andplateELISA,usingmonoxenically-grownEntamoebahistolyticasolublecrudeantigen,fordetectingantiamoebicantibodiesinserumsamplesfromsuspectedamoebicpatientsandhealthycontrols.MaterialsandMethods:Serafrom18suspectedamoebicpatients,15healthysubjectsand13patientswithotherparasiticdiseases(leishmaniasis,hydatidosis,ascariasisandtrichostrongyloidiasis)wereexaminedforthepresenceofspecificEntamoebahistolyticaantibodiesbyELISAanddot-ELISA.BothdotandplateELISAdetectedantiamoebicantibodiesin15(outof18)suspectedamoebic.Results:From15healthycontrols,twoserawerefoundtobepositivebyELISAandoneofthesetwowasalsopositivebydot-ELISA.Inbothassays,nocrossreactionwasfoundwiththeserafromotherparasiticdiseasesincludingleishmaniasis,hydatidosis,ascariasisandstrongyloidiasis.SensitivityoftheELISAsystemwasdeterminedas83.3%(95%CI:57.7-95.6%)andthespecificitywasfoundtobe92.5%(95%CI:74.2-98.7%).Positiveandnegativepredictivevaluesofthesystemwere88%and89%respectively.Ontheotherhand,sensitivityofthedot-ELISAwasdeterminedas83.3%(95%CI:57.7-95.6%)andthespecificitywasfoundtobe96.3%(95%CI:79-99.8%).PositiveandnegativepredictivevaluesofdotELISAwere93.7%and89.6%respectively.Conclusion:Whilethesensitivityofbothassayswereequal(83.3%),dot-ELISAwasfoundtobemorespecific(96.3%)indetectingantiamoebicantibodiescomparedtoPlateELISA(92.5%).SeeingthatthedotELISAissimpler,rapid,inexpensive,equallysensitiveandmorespecificascomparedtoplateELISA,itsuseinserodiagnosisofamoebiasiscanberecommended.
