[摘要] Inthisstudy,wesoughttoinvestigatethepatternofcellularglycoproteinsduringyeast-to-filamentgrowthtransitioninCandidaalbicans,invitro.BymeanofglycoproteinextractionwithConcanavalinA-Sepharose,andWesternBlotanalysiswithperoxidase-labeledlectins(concanavalinA,wheatgermagglutinin),wehavecharacterizedtwoglycoproteins(52kDaand42kDa)whosepatternisincreasedinthefilamentousformofC.albicans.AnalysisofsubcellularfractionsofC.albicansshowedthatthe52/42kDaglycoproteinsarelocatedinthesolublefraction.invitrotreatmentofconcanavalinA-Sepharoseextracted52/42kDaglycoproteinswiththepeptideN-linkedglycosidaseFshowedthatthe52kDaproteinishighlyN-glycosylatedandmannose0-glycosylated,whereasthe42kDaisN-glycosylated.Regulationofsynthesisand/orglycosylationofthe52/42kDaglycoproteinscouldbeassociatedwithyeast-to-filamentgrowthtransitionofC.albicans.
