IgG antibody patterns in aqueous humor of patients with primaryopen angle glaucoma and pseudoexfoliation glaucoma
[摘要] Purpose: There has been growing body of evidence indicating animmunological component in the pathogenesis of glaucoma. Several studiesfound specific antibodies in sera of glaucoma patients. Recently, wedetected antibody profiles in aqueous humor of patients with normaltension glaucoma that showed significant differences when compared tocontrol subjects. The aim of this study was to compare the IgG antibodypatterns against retinal antigens in aqueous humor of patients withprimary open angle glaucoma, pseudoexfoliation glaucoma, and controlsubjects then to identify some of the important biomarkers of thisstudy.Methods: Aqueous humor of 44 patients was analyzed: 15 patients withprimary open angle glaucoma (POAG), 14 patients with pseudoexfoliationglaucoma (PEX), and 15 control subjects (CO). The aqueous was testedagainst western blots of bovine retinal antigens. The IgG antibodypatterns were analyzed by multivariate statistical techniques. Some ofthe important biomarkers were identified via mass spectrometry(Maldi-TOFTOF).Results: All patients showed complex patterns of IgG antibodiesagainst retinal antigens. The discriminant analysis revealed astatistically significant difference between the antibody profiles ofthe POAG and the CO group (p=0.00018). There was also a statisticallysignificant difference between the antibody profiles of the PEX and theCO group (p=0.0013). Not only were up-regulations in the glaucoma groupsobserved, compared to controls, but also down-regulations. There was nosignificant difference between the antibody patterns of the POAG and PEXgroup. The identified biomarkers included heat shock protein 27,α-enolase, actin, and GAPDH.Conclusions: We could show significant differences between the IgGantibody profiles of the glaucoma groups (PEX and POAG) and controls.The analysis of intraocular antibodies could provide further hints forautoimmune involvement in glaucoma.
