The Aggregation of γ-Myeloma Proteins

[摘要] The degree of aggregation of 18 isolated γG-myelomas was evaluated by reaction with rheumatoid factor, inactivation of complement and ultracentrifugal analysis. Eleven of 18 myelomas were not aggregated by heat or mercaptoethanol-urea treatment. Of the remaining seven, four showed virtually complete aggregation and three appeared to be partially aggregated. Preparations of normal γG-globulin were approximately 20% aggregated.In evaluating the degree of aggregation, the methods of ultracentrifugation and precipitation with rheumatoid factors of broad specificity correlated well. Since many isolated myelomas were anticomplementary prior to heating and varied in their ability to inactivate complement after heating, complement inactivation was a poorer method for evaluating aggregation.
