Microspectrophotometric Determination of DNA Quantities and Polypliody in Fraxinus america L.
[摘要] Quantitative determination of DNA amounts per cell for66 Fraxinus americana trees, from 22 locations, was accomplishedby Feulgen microspectrophotometric methods. Measurementsof arbitrary photometric units of relative absorbanceof the DNA-Feulgen dye complex were adjusted to estimates ofactual DNA amounts by the use of chicken erythrocyte nuclei.A direct correlation was found between observed DNA amountsand diploid, tetraploid and hexaploid levels. A deviationof observed ploidy DNA values from a direct multiplicationfactor suggests that aneuploidy may be operating in thepopulation adaptation processes. South of 40° north latitudeboth diploid and polyploid individuals were representedwhile north of 40° north latitude only diploid individualswere found.
