New Automated Method for Measuring Thiamine (Vitamin B1) in Urine

[摘要] A new automated method was developed to eliminate the time-consuming steps of the manual procedure for the measurement of thiamine (vitamin B1) in urine. A Technicon Pump II simultaneously pumped 23 samples through chromatographic columns of "Decalso," washed out the interfering substances, and eluted thiamine. The automated procedure for determining thiamine with a Technicon AutoAnalyzer system involved the oxidation of thiamine to thiochrome, extraction into isobutanol and fluorescence measurements. Results obtained by the automated method correlated well with those obtained by the manual procedure. Recoveries of added thiamine were satisfactory and the results reproducible. This method was used successfully in the analysis of more than 10,000 urine samples for the Canadian Nutrition Survey.
