FUSIONOF RAW 264.7 MACROPHAGE CELLSONMICRO SCALE FINE PILLAR PATTERN S
[摘要] Macrophage dependent immune response to the implant surface sometimes results infailure of implant. M acrophage behaviors are sensitive to the topological factors involvingmicro/nano patterns on the implant s urface . However , further investigations aboutmacrophage behaviors toward the fine micro/nano patterns on the implant surface arerequired. In the present study, we investigated the effect s of fine micro scale pillar pattern sfound on the surface of implants on macrophage fusion. Micro pillar s with dia meters of 2µm, 1 µm, and 500 nm and a height of 500 nm were fabricated by nanoimprinting on acyclo olefin polymer (COP) film. RAW 264.7 macrophages attachment assay show s that 1µm and 500 nm pillar s improved cell attachment than the planar surface. Macrophagefusion assay shows that the number of multi nucleated cell s on 1 µm and 500 nm pillar swas approximately 1. 4 to 1.5 times higher than that on the planar surface . However, 2µm pillar was ineffective in both the assays Interestingly, the expression of F actin andvinculin in mononuclear and multinucleated cells on the pillar s appeared ring shaped alongthe circumference of pillars , indicating frustrated phagocytosis . We suggest that finemicro patterning of the substrate could be one of th e factor s that control cell attachmentand fusion of RAW 264.7 cells.
