Comparison and improvement of the existing methods for the determination of aflatoxins in human serum by LC-MS/MS
[摘要] AflatoxinsaretoxicmetabolitesofsomeAspergillusflavus,A.parasiticusandA.nomiusstrainsthatoccurinmanyfoodsandfeeds.AflatoxinB1isthemostabundantandtoxicmemberofthefamily,anditisalsothemostpotenthepatocarcinogenknown.InordertoestimatethepotentialhumanhealthriskofaflatoxinB1,itisusefultomeasureitsbloodconcentration.Inthisworkarapidliquidchromatographycoupledtomassspectrometry(LC-MS/MS)methodforthesimultaneousdeterminationofaflatoxinsB1,B2,G1andG2anditsmetabolites,aflatoxinsM1andM2inhumanserum,isproposed.Smallserumvolumeshavebeenusedandtheextractshavebeenobtainedwithoutanyclean-upprocedure.TheisolationoftheaflatoxinsfromhumanserumwasachievedusingsubsequentpH-controlledsolventextractionssteps.Recoveryratesintherangebetween31and98%wereachievedintheserum,whilethelimitsofdetectionandquantificationfortheproposedmethodwere0.5ngmL−1and1.0ngmL−1,respectively.Thevalidationofthemethodhasbeendemonstratedbyanalysingspikedandnaturallycontaminatedbloodserum...
