Comparative investigation of PdR by usual and ultrafine atomic force microscopy
[摘要] Atomicforcemicroscopyisoneofthemostperspectivemethodsfordeterminationofthestructureofproteinsandtheircomplexes.Theverticalresolutionofthismethodisabout0.1nm,whichisclosetoX-rayresolution.Atthesametime,thelateralresolution,determiningbybroadeningeffectofastandardAFMprobe,isnotveryhigh—about20–50nm,dependingonprobegeometry.Naturally,theprobetipbroadeningeffectleadstosubstantialenhancementofmeasuredproteinvolume.Inthisstudy,acomparativeanalysisofsizesoftheproteinputidaredoxinreductase(PdR)obtainedbytheuseoftwoAFMprobetypes,standardandsupersharp,wasundertaken.UsageofstandardprobesenabledtocorrectlymeasuretheheightofPdRwhilethevolumeofthisproteinwasmeasuredwithconsiderably(morethanoneorder)enhancement.ItwasshownthatapplicationofsupersharpAFMprobesresultsintheloweringofmeasuredproteinheight;atthesametime,themeasuredproteinvolumeismoreexactandappearstobeclosetoRSAdata.Therefore,toobtainexactdataonproteinvolumeandheight,thesetwoparametersshouldbemeasuredbyuseofbothsupersharpprobesandstandardgeometryprobes...
