Determination of phosphatidylcholine in soybean lecithin samples by high performance liquid chromatography on titania
[摘要] Highperformanceliquidchromatography(HPLC)ontitaniacolumnwasperformedforphosphatidylcholine(PC)separationandanalysis.TofullyunderstandtheretentionmechanismofPContitania,theeffectofmobilephasetype,%acetonitrile(%ACN),phosphatebufferconcentration,bufferpHandflowratewereallstudied.SeparationdependedontheeluentconditionsandPCretentionwasattributedtothecooperationofhydrophilicinteractionandligandexchange.PCinsoybeanlecithinproductswascompletelyseparatedfromothercomponentswithin6minandquantitativeanalysiswasachievedbyanexternalstandardmethod,withacorrelationcoefficient(R2)of0.9947.ThemethodprecisionanddetectionlimitofPCwere3.40%and0.2μg,respectively,withtherecoveryrangingfrom91%to101%.Moreover,PCandglycerolmonostearatestandardmixturewassuccessfullyisolatedbytheproposedmethod,whichunderlinedtheselectiveaffinityoftitaniaforphosphorylatedlipids.Withthesuperiorityofsimplicity,rapidityandaccuracy,thisHPLCmethodbasedontitaniaisacompetentmethodforthedeterminationofPCinlecithinsampleswithprospectsinthefieldoforgano-phosphatesanalysis...
