Pinpointing oxidative modifications in proteins—recent advances in analytical methods
[摘要] Oxidationisoneofthekeydegradationpathwaysinproteinsandisofrelevancetoanalyzeinawidevarietyofresearchdisciplines.Thetypesofdifferentmodificationsoccurringduringoxidationbyreactiveoxygenspecies(ROS)arealmostasmanyasthenumberofavailableanalyticalmethods.Proteinoxidationinbiologicalsampleshastraditionallybeenanalyzedbyvariousproteincarbonylassays,sometimescombinedwithmassspectrometry(MS)foridentificationofthecarbonylatedproteins.MSisnowincreasinglyusedalsoforthedeterminationoftheexactpositionoftheoxidationintheaminoacidsequence,anapproachthatisaidedbyrecentdevelopmentswithinthefieldofproteomics.ThefollowingreviewsummarizestheeffectsofROSonproteins,describesmethodsforlabelingandseparationofoxidizedproteinsincomplexmixtures,andprovidesinsightintovariousMS-basedmethodstolocalizethemodificationswithintheproteinprimarystructure.Pitfallswiththedifferenttechniquesaregiven,aswellasexamplesfromvariousapplicationswithinbiologicalstudies,proteintherapeutics,plantscience,thefoodindustryandindustrialbiotechnology...
