Development of a screening method for the detection of analogues of sildenafil and vardenafil by the use of liquid chromatograph coupled with triple quadrupole linear ion trap mass spectrometer

[摘要] Sildenafilandvardenafilarewesterndrugsusedinthetreatmentofmaleerectiledysfunction(ED).PrescriptionisrequiredtousethesedrugsinmanycountriesincludingHongKong.Recently,healthsupplementsandChineseherbaldrugsclaimingtohaveimprovementinmalepotencywerefoundtobeadulteratedwiththesedrugs.Evenworse,someoftheseproductswerefoundtocontainanaloguesofEDdrugs.Forregulatorypurposes,drugenforcementagenciesoftenuseanalyticaltechniquessuchasHPLC-UVorLC-MS/MSinscheduledmultiplereactionmonitoring(MRM)modeforroutinescreeningofwesterndrugingredients.However,druganaloguesareexpectedtohavedifferentelutionprofilesandtheionsproducedbydruganalogueswouldbedifferent,makingthemtransparentinscheduledMRMconditions.Totackletheproblem,anacquisitionmethodthatcombinedprecursorionscan(PIS)withinformationdependentacquisition(IDA)followedbyenhancedproduction(EPI)scanwasdeveloped.Resultssuggestedthatsuchanapproachcouldprovideaverypowerfultoolinthedetectionofanaloguesofsildenafilandvardenafil...
