Characterization of flavone glycosides and aglycones in Cephalotaxus sinensis by HPLC-DAD-MS
[摘要] Highperformanceliquidchromatographydiodearraydetectorelectrosprayionizationmassspectrometry(HPLC-DAD-ESI-MS)wasutilizedtoseparateandidentifyflavoneglycosidesandaglyconesinCephalotaxussinensis,anaturalplantwithananti-hyperglycemiceffect.TheHPLC-DAD-ESI-MSsystemcouldprovideagreatamountofinformationonthestructureofflavoneaglyconesandglycosidesinC.sinensiswithouttime-consumingpre-separationorpurificationprocedures.Atotalofsevenflavoneglycosidesandtwoflavoneaglyconeswereunambiguouslyidentifiedbasedontheirretentiontimes,ultravioletspectra,molecularweightsandMS/MSspectra.It'sinterestingthatfive5-O-flavoneglycosideswerefoundinC.sinensissince5-O-flavoneglycosidesarerareanddifficulttoforminthenaturalplant.Amongthem,quercitrinandapigenin-5-O-β-D-glucopyranosidewereisolatedfromthisgenusforthefirsttime.Anovelflavoneglycoside,chryoseriol-5-O-[α-L-rhamnopyranosyl-(1→4)-β-D-6-O-acetylglucopyranoside],wasisolatedforthefirsttimeandidentifiedbytheultravioletspectrum,infraredspectrum,massspectrumandnuclearmagneticresonancespectrum.Inaddition,theproposedfragmentpathwayofthenovelflavoneglycosidewasconcludedinthenegativeionizationmodebyHPLC-ESI-MS/MS...
