Monolithic porous layer open tubular (monoPLOT) columns for low pressure liquid chromatography of proteins

[摘要] Glycidylmethacrylate–ethylenedimethacrylate(GMA-co-EDMA)basedmonolithicporouslayeropentubular(monoPLOT)columns(0.05mmI.D.,monolithiclayerthickness≈5µm)havebeenfabricatedusinganautomatedcolumnscanningtechnique,providingUVpolymerisationat365nm.Columnswerechemicallymodifiedtoobtaindesireddiolgroupsonthesurface,andthelongitudinalhomogeneityofthestationaryphasewasprofiledusingscanningcapacitivelycoupledcontactlessconductivitydetector(sC4D),beforeandaftersuchmodification.Usingtheautomatedscanningpolymerisationtechnique,column-to-columnproductionreproducibility,includinglongitudinalphasethickness,waswithin±5%RSD.Thepreparedcolumnsweretestedtoevaluatetheirliquidchromatographicstationaryphaseselectivity,efficiencyandreproducibility,withaseriesoftestproteinmixtures.Underoptimisedgradientconditions,theseparationofupto8proteinswasdemonstratedontheopentubularcolumn(510×0.05mmI.D.),withacolumnpressuredropof<1.5MPa...
