[摘要] Hypoxiaisanimportantfeatureofinflamedtissue,suchastheRAjoint.Activatedmonocytes/macrophagesandendothelialcellsplayapivotalroleinthepathogenesisofRA,implicatedinthemechanismofinflammationanderosion.Duringdevelopment,myeloidprogenitorcellssequentiallygiverisetomonoblasts,promonocytes,andmonocytesthatarereleasedfromthebonemarrowintothebloodstream.Afterextravasation,monocytesdifferentiateintolong‐lived,tissue‐specificmacrophagesorDCs.Theeffectofdifferentoxygenconcentrationsexperiencedbythesecellsduringmaturationrepresentsanovelaspectofthisdevelopmentalprocess.Ininflamedjointtissue,themicrovasculararchitectureishighlydysregulated;thus,efficiencyofoxygensupplytothesynoviumispoor.Therefore,invadingcellsmustadaptinstantaneouslytochangesintheoxygenlevelofthemicroenvironment.Angiogenesisisanearlyeventintheinflammatoryjoint,whichisimportantinenablingactivatedmonocytestoenterviaendothelialcellsbyactiverecruitmenttoexpandthesynoviumintoa“pannus”,resultingincartilagedegradationandbonedestruction.Theincreasedmetabolicturnoveroftheexpandingsynovialpannusoutpacesthedysfunctionalvascularsupply,resultinginhypoxia.Theabnormalbioenergeticsofthemicroenvironmentfurtherpromotessynovialcellinvasiveness.InRA,jointhypoxiarepresentsapotentialthreattocellfunctionandsurvival.Notably,oxygenavailabilityisacrucialparameterinthecellularenergymetabolism,itselfanimportantfactorindeterminingthefunctionofimmunecells...
