The effect of adenovirus‐specific antibodies on adenoviral vector–induced, transgene product–specific T cell responses
[摘要] Inthisstudy,wetestedtheeffectofneutralizingAbstodifferentserotypesofE1‐deletedAdvectorsontheimmunogenicityofthehomologousAdvectororavectorderivedfromaheterologousserotype.Ourresultsshowedthat,asexpected,evenlowtitersofpassivelytransferredneutralizingAbssignificantlyreducedthehomologousvectors'abilitytoelicittransgene‐specificCD8+Tcellresponses.Inaddition,Abschangedthefateoftransgeneproduct–specificCD8+Tcellsbypromotingtheirtransitionintothecentralmemorycellpool,whichresultedinmarkedlyenhancedexpansionoftransgeneproduct–specificCD8+TcellsafteraboostwithaheterologousAdvector.Non‐neutralizingAbsspecifictoadistinctAdserotypehadnoeffectonthemagnitudeoftransgeneproduct‐specificCD8+TcellsinducedbyaheterologousAdvector,nordidsuchAbspromoteinductionofmorerestingmemoryCD8+Tcells.TheseresultsshowthatAbstoanAdvaccinecarrieraffectnotonlythemagnitudebutalsotheprofileofavector‐inducedCD8+Tcellresponse...
