Interaction of members of the heat shock protein‐70 family with the macrophage mannose receptor
[摘要] ThemacrophageMRhasbeenthesubjectofinvestigationforover20years,andseveralimportantphysiologicalfunctionshavebeendescribed.However,themolecularmechanismsthatregulateMRsignalingandtraffickingduringtheseprocessesstillremainelusive.ThefocusofthecurrentpaperwastoidentifypotentialcellularMR‐interactingproteins.AninitialscreenofbindingproteinsinMR‐expressingcellswasperformedusingcoimmunoprecipitation,followedbyidentificationofmatchingpeptidesequencesusingproteomicsandMS.Themajorclassofbindingproteinsidentifiedbelongedtotheheatshockfamilyofproteins.ThespecificinteractionoftheMRwithHSP70familymemberswasvalidatedbyWesternblotanalysis,ligandbindingassays,andintracellularcolocalizationusingconfocalmicroscopy.AdditionalstudiesindicatedthatinhibitionoftheHSPBiPbytreatmentofcellswithEGCGreducedBiPinteractionwithandsurfaceexpressionoftheMR.StudiesofpossiblemotifswithinthecytoplasmictailofthereceptorsuggestedthatajuxtamembranedibasicsequencemaycontributetotheinteractionwithBiP.ThesefindingssuggestthatthemolecularassociationoftheMRwithHSP70familymembersviathereceptorcytoplasmictailmaycontributetoMRtraffickinginmacrophages...
