NK cells are necessary for recovery of corneal CD11c+ dendritic cells after epithelial abrasion injury
[摘要] MechanismscontrollingCD11c+MHCII+DCsduringcornealepithelialwoundhealingwereinvestigatedinamurinemodelofcornealabrasion.SelectivedepletionofNKp46+CD3−NKcellsthatnormallymigrateintothecorneaafterepithelialabrasionresultedin>85%reductionoftheepithelialCD11c+MHCII+DCs,normallypresentduringandafterepithelialwoundclosure.Transfer(i.v.)ofspleenNKcellsintoNKcell‐depletedmicesignificantlyrestoredlevelsofcornealepithelialDCs(P<0.01).ImmigratedNKcellswerepredominatelypositiveforIFN‐γ,andtopicalcornealanti‐IFN‐γreducedepithelialDCsby79%(P<0.01).IFN‐γ−/−micehad69%fewerDCsthanWTcontrols(P<0.01),andtopicalrIFN‐γappliedtoNKcell‐depletedcorneasincreasedepithelialDCssignificantly(P<0.01).ThecontributionofICAM‐1,anadhesionmoleculeinvolvedinleukocytemigration,expressedonhealingcornealepithelium,wasevaluated.ICAM‐1−/−miceexhibited>70%reductioninepithelialDCrecoveryinthefirst48hafterepithelialabrasion(P<0.01).TheseinterventionsrevealanearlyturnoverofDCsintheepitheliumafterinjury,andICAM‐1,NKcells,andIFN‐γarenecessaryfortheimmigrationphaseofthisturnover...
