Enhancement of humoral immunity by the type II heat‐labile enterotoxin LT‐IIb is dependent upon IL‐6 and neutrophils
[摘要] LT‐IIb,atypeIIheat‐labileenterotoxinproducedbyEscherichiacoli,isapotentintradermaladjuvantthatenhancesimmuneresponsestocoadministeredantigens.Althoughtheimmunemechanismsthatpromotethisaugmentedimmuneresponsehavenotbeenwelldefined,priorintradermalimmunizationexperimentssuggestedthatearlycellularandimmunomodulatoryeventsatthesiteofimmunizationmodulatedtheaugmentationofantigen‐specificimmuneresponsesbyLT‐IIb.Toinvestigatethathypothesis,micewereintradermallyimmunizedwitharecombinantricinvaccine,aprospectivetoxinsubunitantigen,inthepresenceandabsenceofLT‐IIb.Analysisoftissue‐fluidcollection,coupledwithhistologicsectionsfromthesiteofintradermalimmunization,revealedthatasingledoseofLT‐IIbinducedlocalproductionofinterleukin6andpromotedaregionalinfiltrationofneutrophils.TheadjuvanteffectsofLT‐IIbwereabrogatedininterleukin6–deficientmiceandwhenmiceweredepletedofneutrophilsbypretreatmentwithanti‐Ly6G.Overall,thesedatafirmlydemonstratedthatLT‐IIb,whenusedasanintradermaladjuvant,recruitsneutrophilsandisapotentrapidinducerofinterleukin6...
