[摘要] In opposition to the old dogma of the free diffusion of cAMP within the cell, today, experimental results support the existence of microdomains of cAMP, maintained by physical barriers and restricted diffusion of the nucleotide. Compartmentalization of the signal is mainly due to the family of anchor proteins AKAP. Originally described as proteins anchoring PKA, the AKAPS have the ability to orchestrate the assembly of multiprotein complexes with other proteins involved in the cAMP signaling pathway, such as ACs, PDEs and Epac. The presence of PDEs in these signaling microdomains is of fundamental importance because they restrict the free diffusion of the messenger and limite its availability. Thus, compartmentalization regulates the location, duration and amplitude of the signal, essential requirements for an efficient transduction of the stimulus.
