Adiponectin deficiency-induced diabetes increases TNFα and FFA viadownregulation of PPARα
[摘要] Expression of peroxisome proliferator-activated receptor (PPAR) α was investigated inadiponectin knockout mice to elucidate the relationship between PPARα and adiponectindeficiency-induced diabetes. Adiponectin knockout (Adpâ/â) mice were generatedby gene targeting. Glucose tolerance test (GTT), insulin tolerance test (ITT), and organsampling were performed in Adpâ/â mice at the age of 10 weeks. PPARα, insulin,triglyceride, free fatty acid (FFA), and tumor necrosis factor α (TNFα) were analyzed fromthe sampled organs. Adpâ/â mice showed impaired glucose tolerance and insulinresistance. Additionally, PPARα levels were decreased and plasma concentration oftriglyceride, FFA and TNFα were increased. These data may indicate that insulin resistancein Adpâ/â mice is likely caused by an increase in concentrations of TNFα andFFA via downregulation of PPARα.
