Rat resident testicular macrophages have an alternatively activated phenotype and constitutively produce interleukin‐10 in vitro
[摘要] Theabilityoftherodenttestistotolerategraftalloantigensandspermatogeniccellautoantigensiswellknown.Themechanismsunderlyingthis“immuneprivilege”arepoorlyunderstood,butthenumerousresidentTMshavebeenimplicated.AlthoughithasbeenassumedthatTMsdisplayaphenotypeconsistentwithimmuneprivilege,thishasnotbeenformallyestablished.Consequently,TMswereisolatedfromadultratsandculturedunderbasalconditionsandfollowingstimulationwithLPSandIFN‐γ(classicalactivation)orIL‐4(alternativeactivation).BMMsmaturedinvitrowereusedascontrol.Expressionoftheclassical(proinflammatory)activationmarkersTNF‐α,IL‐1β,iNOS,IL‐6,RANTES,IL‐12p40,andSOCS3andalternative(immunoregulatory)activationmarkersIL‐10,TGF‐β1,CXCL2,andSOCS1wasmeasuredbyQPCRorELISA.Inculture,TMswerecharacterizedbypoorexpressionofclassicalactivationgenesandTGF‐β1butconstitutivelyhighIL‐10productionandreducedcostimulatoryactivityinapolyclonalTcellactivationassay.ThispatternofgeneexpressionwasassociatedwithTMsexpressingthescavengerreceptorCD163,whichischaracteristicoftissueresidentmacrophagesandalternativeactivation.Bycontrast,CD163‐negativeTMsdisplayedreducedinflammatorygeneexpressionbutdidnotconstitutivelyproduceIL‐10.Thesedataindicatethatundertheinfluenceofthetesticularenvironment,macrophagesadoptanalternativelyactivatedphenotype,involvingreducedcapacityforproinflammatorygeneexpression,constitutiveIL‐10production,andimpairedabilitytosupportTcellactivation,consistentwitharoleinmaintainingtesticularimmuneprivilege...
