Effective T helper cell responses against retroviruses: are all clonotypes equal?
[摘要] ThecriticalimportanceofCD4+TcellsincoordinatinginnateandadaptiveimmuneresponsesisevidencedbythesusceptibilitytovariouspathogenicandopportunisticinfectionsthatarisesfromprimaryoracquiredCD4+Tcellimmunodeficiency,suchasfollowingHIV‐1infection.However,despitetheclearlydefinedrolesofcytotoxicCD8+Tcellsandantibodiesinhostprotectionfromretroviruses,theabilityofCD4+Tcellstoexertasimilarfunctionremainsunclear.RecentstudiesinvarioussettingshavedrawnattentiontothecomplexityoftheTcellresponsewithinandbetweenindividuals.DistinctTCRclonotypeswithinanindividualdiffersubstantiallyintheirresponsetothesameepitope.Functionallysimilar,“public”TCRclonotypescanalsodominatetheresponseofdifferentindividuals.TCRaffinityforantigendirectlyinfluencesexpansionanddifferentiationofrespondingTcells,alsolikelyaffectingtheirultimateprotectivecapacity.WiththisincreasingunderstandingoftheparametersthatdeterminethemagnitudeandeffectortypeoftheTcellresponse,wearenowbetterequippedtoaddresstheprotectivecapacityagainstretrovirusesofCD4+Tcellclonotypesinducedbynaturalinfectionorvaccination...
