HIV infection of macrophages is enhanced in the presence of increased expression of CD163 induced by substance P
[摘要] ActivationofNK1RbySPcontributestoincreasedHIV‐1infectioninmacrophages.ThescavengerreceptorCD163isexpressedoncellsofmonocyte‐macrophageorigin.OurmaingoalwastodetermineifthereisinterplayamongSP,CD163expression,andHIVinfectioninmacrophages.WeshowedthatSPtriggersintracellularcalciumelevationandincreasedCD163expressioninhumanmonocytesinatime‐andconcentration‐dependentmanner.TheroleofCD163onHIVinfectionwasexaminedbyRT‐PCRinsortedmonocytes(CD163lowandCD163high)andinmacrophageshavingCD163knockeddownusingsiRNA.WefoundthattheproductivityofHIVinfectionwashigherinCD163highcells.Additionally,inmacrophageswithCD163expressionknockeddown,wefoundasignificantdecreaseofHIVinfection.Furthermore,Hb‐Hpcomplexes,whichfunctionasanendogenousligandforCD163,decreasedHIVinfectioninmacrophagesinadose‐dependentmanner.Thus,wedemonstratethatSPinduceshigherlevelsofCD163inmonocytesandthathighexpressionofCD163isassociatedwithincreasesHIVinfectioninmacrophages.Thus,inadditiontobeingaprognosticmarkerofHIVinfection,theexpressionofCD163onmacrophagesmaybecriticalinHIVimmunopathogenesis...
