Complement modulation of T cell immune responses during homeostasis and disease
[摘要] Thecomplementsystemisanancientandcriticaleffectormechanismoftheinnateimmunesystemasitsenses,kills,andclearsinfectiousand/ordangerousparticlesandalertstheimmunesystemtothepresenceoftheinfectionand/ordanger.Interestingly,anincreasingnumberofreportshavedemonstratedaclearroleforcomplementintheadaptiveimmunesystemaswell.Ofnote,anumberofrecentstudieshaveidentifiedpreviouslyunknownrolesforcomplementproteins,receptors,andregulatorsinTcellfunction.Here,wewillreviewrecentdatademonstratingtheinfluenceofcomplementproteinsC1q,C3b/iC3b,C3a(andC3aR),andC5a(andC5aR)andcomplementregulatorsDAF(CD55)andCD46(MCP)onTcellfunctionduringhomeostasisanddisease.AlthoughnewconceptsarebeginningtoemergeinthefieldofcomplementregulationofTcellfunction,futureexperimentsshouldfocusonwhethercomplementisinteractingdirectlywiththeTcellorishavinganindirecteffectonTcellfunctionviaAPCs,thecytokinemilieu,ordownstreamcomplementactivationproducts.Importantly,theidentificationofthepivotalmolecularpathwaysinthehumansystemswillbebeneficialinthetranslationofconceptsderivedfrommodelsystemstotherapeutictargetingfortreatmentofhumandisorders...
