CD14hiHLA‐DRdim macrophages, with a resemblance to classical blood monocytes, dominate inflamed mucosa in Crohnˈs disease
[摘要] IntestinalMϕplayanimportantroleinmaintainingguthomeostasis.However,littleisknownaboutthesecells,theirprecursors,andtheirroleinintestinalinflammation.Here,wecharacterizetheCD14+mononuclearcellpopulationsinintestinalmucosaandbloodinpatientswithCD.AmongtheLPCD14+Mϕ,weidentifiedthreedistinctHLA‐DR+‐expressingsubsets.Comparedwithuninflamed,inflamedmucosacontainedamarkedincreaseintheproportionoftheCD14hiHLA‐DRdimcellularsubset.ThissubsetresembledtheclassicalbloodmonocyteswithlowCD16,HLA‐DR,andCX3CR1expression.ClassicalmonocytesmigratedefficientlytowardCCL2andreleasedthehighestlevelsofMMP‐1andproinflammatorycytokineswhenstimulatedwithimmunecomplexesorLPS.OurfindingsstronglysuggestthatitistheclassicalandnottheintermediateornonclassicalmonocytesthataretheprecursorstothedominatingintestinalCD14hiHLA‐DRdimsubset.ThisenhancesourunderstandingofCDpathologyandmayprovidenewoptionsintreatment...
