Characterization of tetraspanins CD9, CD53, CD63, and CD81 in monocytes and macrophages in HIV‐1 infection
[摘要] Tetraspaninsareafamilyofmembrane‐organizingproteinsthatmediatediversefunctions.Littleisknownoftheirexpressionorfunctioninmyeloidcells.Here,expressionofCD9,CD53,CD63,andCD81,tetraspaninsthathavebeenimplicatedinHIV‐1pathogenesis,werecharacterizedinnormalmonocytesubsets,inMDM,andinHIV‐1‐infecteddonors.Weshowthattetraspaninsareexpresseddifferentiallybymonocytesubsets,withhigherCD9andCD63andlowerCD53andCD81levelsonCD14++CD16−monocytescomparedwithCD14++CD16+andCD14+CD16++subsets.MaturationofmonocytesresultedinincreasedCD9expressionandapparentrelocationofCD63andCD53fromsurfacetointracellularmembranes.Expressionwasmodulatedbycytokines,andCD9wasamarkerofanti‐inflammatoryandCD53amarkerofproinflammatoryMDM.TetraspaninexpressiononmonocytesubsetsfromHIV‐1‐infecteddonorsreceivingantiretroviraltherapywasunchangedcomparedwiththatinuninfecteddonors.However,CD53expressionwasinverselycorrelatedwithviralloadinHIV‐1‐infecteddonorsnotontherapy.ThisstudyisthefirsttocomprehensivelycharacterizetetraspaninexpressiononmonocytesubsetsandmacrophagesinhealthandduringHIV‐1infection.Itdemonstratesregulationoftetraspaninexpressionbycytokines,andCD53expressionasanovelcorrelateofaproinflammatoryphenotype.Thispapercharacterizestetraspaninsinmyeloidcellsandshowsthattetraspaninsareexpresseddifferentiallyinmonocytesubsetsandaremodifiedininflammatoryconditions...
