The roles of complement receptor 3 and Fcγ receptors during Leishmania phagosome maturation
[摘要] Leishmaniaareintracellularparasitesadaptedtosurvivinginmacrophages,whoseprimaryfunctioniseliminationofinvadingpathogens.Leishmaniaentryintohostcellsisreceptor‐mediated.Theseparasitesareabletoengagemultiplehostcell‐surfacereceptors,includingMR,TLRs,CR3,andFcγRs.Here,weinvestigatedtheroleofCR3andFcγRengagementonthematurationofLeishmania‐containingphagosomesusingCD11b−/−andFcγR−/−macrophages,andassessingEEA1andlysosome‐associatedproteinsisnecessaryforthephagosomematurationdelay,characteristicofLeishmaniainfection.Leishmania‐containingphagosomesdonotfusewithlyosomesuntil5hpostinfectioninWTmice.Phagolysosomefusionoccursby1hinCD11bandFcγRcommonchainKOmacrophages,althoughreceptordeficiencydoesnotinfluenceLeishmaniaentryorviability.Wealsoinvestigatedtheinfluenceofserumcomponentsandtheireffectsonphagosomematurationprogression.Opsonizationwithnormalmouseserum,complement‐deficientserum,orserumfromLeishmania‐infectedmiceallinfluencedphagosomematurationprogression.OurresultsindicatethatopsonophagocytosisinfluencesphagosomaltraffickingofLeishmaniawithoutalteringtheintracellularfate...
