Mass isolation of pole cells from Drosophila melanogaster☆
[摘要] Crudecellsuspensionswereobtainedfrom3×106pregastrulastagedembryos.ThesecellsfractionatedintothreebandsonRenografindensitygradients.Usingultrastructuralcharacteristicsforidentification,polecellswerelocalizedinthetwodenserbands.EManalysesalsorevealedastrikingdifferenceinthefrequencyoflipidvacuolesfoundinthepole-cellcytoplasmversusthecytoplasmofotherembryoniccells.Thisdifferencehasenabledustoidentifypolecellsbylightmicroscopyusinganeutrallipidstain.ThroughdetailedanalysesoftheRenografinfractionswiththisstain,wehaveshownthatpolecellsresolveintotwotothreedensityclasses.Evidenceispresentedwhichsuggeststhatthesedensityclassesreflectdifferentdevelopmentalageclasses.Thesizedifferencesbetweenthepolecellsandotherembryoniccellscontainedintheenrichedpole-cellfractionsfromthedensitygradientshasenabledustousesedimentationvelocitycentrifugationforadditionalenrichment.Adistinctlowerbandofcellswasobtainedwhichconsistedof80%polecells.Usingtheseprocedures,1–2×107polecellscanbeobtaineddaily.
