Sequence complexity of the RNA accumulated in oocytes of Arbacia punctulata☆
[摘要] ThesequencecomplexityoftheRNApresentinmatureoocytesoftheseaurchinArbaciapunctulatawasmeasuredbyDNA-RNAhybridization.Smallquantitiesofsinglecopy[3H]DNAwerehybridizedwithlargeexcessesofRNAandtheamountof[3H]DNA-RNAhybridsformedwasassayedbyhydroxyapatite.Atterminationofthereaction,whichfollowspseudo-first-orderkinetics,2.6%ofthereactablesinglecopy[3H]DNAmassispresentinRNA-DNAhybrids.ThisDNAwasisolated,showntoberehybridizabletoRNAand,withintherangeofdetectability,toconsistsolelyofnonrepetitivesequences.Assumingasymmetrictranscription,5.2%ofthegenomicsinglecopysequence,or3.0×107nucleotides,isrepresentedinoocyteRNA.Thekineticsofhybridizationindicatethatthesesequencesareincludedinapproximately1–2%oftheoocyteRNA.
