RAGE regulation and signaling in inflammation and beyond
[摘要] RAGEisakeymoleculeintheonsetandsustainmentoftheinflammatoryresponse.NewstudiesindicatethatRAGEmightrepresentanewlinkbetweentheinnateandadaptiveimmunesystem.RAGEbelongstothesuperfamilyofIgcell‐surfacereceptorsandisexpressedonalltypesofleukocytespromotingactivation,migration,ormaturationofthedifferentcells.RAGEexpressionisprominentontheactivatedendothelium,whereitmediatesleukocyteadhesionandtransmigration.Moreover,proinflammatorymoleculesreleasedfromtheinflamedorinjuredvascularsysteminducemigrationandproliferationofSMCs.RAGEbindsalargenumberofdifferentligandsandisthereforeconsideredasaPRR,recognizingastructuralmotifratherthanaspecificligand.Inthisreview,wesummarizethecurrentknowledgeaboutthesignalingpathwaysactivatedinthedifferentcelltypesanddiscussapotentialactivationmechanismofRAGE,aswellasputativeoptionsfortherapeuticintervention...
