Exogenous application of hydrogen sulfide donor attenuates inflammatory reactions through the L‐selectin‐involved pathway in the cutaneous reverse passive Arthus reaction
[摘要] H2Shasbeenhighlightedrecentlyasanendogenous,gaseoussignalingmolecule,especiallyininflammations.ThedepositionofICinducesanacuteinflammatoryresponsewithtissueinjury.ToassesstherolesofH2SintheIC‐induceddiseases,thecutaneous,reversepassiveArthusreactionwasconductedusingNaHSasaH2Sdonor.Furthermore,weconductedsimilarexperimentsusingselectin−/−micetodeterminetheinvolvementofselectinmoleculesintheH2S‐mediatedpathway.ExogenousapplicationofNaHSdramaticallyattenuatedinflammatoryreactionsinWTmiceassociatedwithArthusreaction.Namely,mRNAexpressionsofTNF‐α,IFN‐γ,andneutrophilnumberswerereducedsignificantlyinthelesionalskinsofNaHS‐treatedWTmicerelativetountreatedones.NaHStreatmentsignificantlyreducedthesethreeparametersinthelesionalskinsofE‐andP‐selectin−/−micebutnotinthoseofL‐selectin−/−mice.QuitesimilarresultswereobtainedintheblockingstudyusingWTmiceinjectedwithmAbtoE‐,P‐,andL‐selectin.OurresultsindicatedthattheexogenousapplicationofNaHSattenuatesinflammatoryresponsesinreversepassiveArthusreactionthroughaL‐selectin‐involvedpathwaybutnotthroughE‐orP‐selectinpathways...
