At the Bedside: Innate immunity as an immunotherapy tool for hematological malignancies
[摘要] Theidentificationofananti‐tumoreffectdisplayedbycellsofinnateimmunityhasopenednewscenarios,notonlyinthefieldofallo‐HSCTbutalsofornontransplantedpatientswithhematologicalmalignanciesorsolidtumors.Donor‐derivedNKcellshavebeenshowntocontributetotheeradicationofmalignantcellsafterallo‐HSCT,whenrecipientslackligandsfortheirinhibitoryreceptors.ThesealloreactivedonorNKcellscanalsokillrecipientAPCsandCTLs,thuspreventingtheoccurrenceofGvHDandgraftrejection.TheroleofactivatingreceptorsonthecapacityofNKcellstokillleukemiatargetshasbecomeevidentinthelastyears.Theadoptiveinfusionofexvivo‐activatedNKcellshasbeeninvestigatedrecentlyinPhaseI/IItrialsonpatientswithhematologicalmalignanciesandsolidtumors,withpromisingresults.γδTlymphocytesarealsoabletodisplayanti‐tumoractivity—thisprovidingthebiologicalrationaleforPhaseI/IItrialsinlymphoproliferativedisordersandsolidtumors.AminobisphosphonatesareclinicallyavailablecompoundsabletoboostγδTcellfunction.AsγδTcellsdonotcauseGvHD,theycouldalsobetransducedwithtumor‐associatedchimericantigenreceptorsandsafelyinfusedinallo‐HSCTrecipients.Basicaspectsofinnateimmunityrelevanttothefieldwillbecoveredbyacompanionreviewarticle...
