An optimized disaggregation method for human lung tumors that preserves the phenotype and function of the immune cells
[摘要] Carefulpreparationofhumantissuesisthecornerstoneofobtainingaccuratedatainimmunologicstudies.Despitetheessentialimportanceoftissueprocessingintumorimmunologyandclinicalmedicine,currentmethodsoftissuedisaggregationhavenotbeenrigorouslytestedfordatafidelity.Thus,wecriticallyevaluatedthecurrenttechniquesavailableintheliteraturethatareusedtopreparehumanlungtumorsforimmunologicstudies.WediscoveredthattheseapproachesaresuccessfulatdigestingcellularattachmentsandECMs;however,thesemethodsfrequentlyaltertheimmunecellcompositionand/orexpressionofsurfacemolecules.Wethusdevelopedanovelapproachtopreparehumanlungtumorsforimmunologicstudiesbycombininggentlemechanicalmanipulationwithanoptimizedcocktailofenzymesusedatlowdoses.Thisenzymaticdigestioncocktailoptimizedcellyieldandcellviability,retrievedallmajortumor‐associatedcellpopulations,andmaintainedtheexpressionofcell‐surfacemarkersforlineagedefinitionandinvivoeffectorfunctions.Toourknowledge,wepresentthefirstrigorouslytesteddisaggregationmethoddesignedforhumanlungtumors...
