Platelet‐neutrophil complex formation—a detailed in vitro analysis of murine and human blood samples
[摘要] Plateletsformcomplexeswithneutrophilsduringinflammatoryprocesses.Theseaggregatesmigrateintoaffectedtissuesandalsocirculatewithintheorganism.Severalstudieshaveevaluatedplatelet‐neutrophilcomplexesasamarkerofcardiovasculardiseasesinhumanandmouse.Althoughmultiplepublicationshavereportedplatelet‐neutrophilcomplexcounts,wenoticedthatdifferentmethodswereusedtoanalyzeplatelet‐neutrophilcomplexformation,resultinginsignificantdifferences,eveninbaselinevalues.Weestablishedaprotocolforplatelet‐neutrophilcomplexmeasurementwithflowcytometryinmurineandhumanwholebloodsamples.Invitroplatelet‐neutrophilcomplexformationwasstimulatedwithADPorPMA.Wetestedtheeffectofdifferentsamplepreparationstepsandcytometersettingsonplatelet‐neutrophilcomplexdetectionandnoticedfalse‐positivecountswithincreasingacquisitionspeed.Platelet‐neutrophilcomplexformationdependsonplateletP‐selectinexpression,andantibodyblockingofP‐selectinconsequentlypreventedADP‐inducedplatelet‐neutrophilcomplexformation.Thesefindingsmayhelpgeneratingmorecomparabledataamongdifferentresearchgroupsthatexamineplatelet‐neutrophilcomplexesasamarkerforcardiovasculardiseaseandnoveltherapeuticinterventions...
