New insights on the natural killer cell repertoire from a thorough analysis of cord blood cells
[摘要] AlthoughCBNKcellsarecharacterizedasimmaturelymphocytes,theirimpressiveexpansionandefficientgraft‐versus‐leukemiaresponsehavebeenhighlightedearlyafterUCBT.TobetterevaluatetheirpotentialassourceofeffectiveNKcells,werevisitedthestudyofNKcellrepertoirefromalargecohortofCBsamples.OurstudyshowedthattheCBNKcellrepertoireappearstobeconstructedearly,dependingonKIRgenecontent,butnotontheautologousHLAenvironment.NKG2AwasexpressedonalargeproportionofCBNKcellsthatinverselycorrelatedwithKIR+NKcellfrequency.Self‐HLAclassImolecule–educatedCBKIR+NKcellspresentalowerspontaneouslysisthandotheiradultcounterparts,whichisprobablyrelatedtothelowexpressionofactivatingNKreceptors.WedescribeforthefirsttimeaproliferativeandcytotoxicNKG2C+NKcellsubsetrepresentingmorethan10%ofCBNKcells.NKG2AstronglyinhibitedCBNKcelldegranulation,anditscoexpressiononNKG2C+NKcellsmaycontributetolimitingtheiractivation.Overall,theCBNKcellrepertoireisconstructedearlyandharborsnumerousfunctionalabilitiessharedbyadultNKcells.Inaddition,theirnaïveviralstatusandfastexpansionconfernumerousadvantagesinimmunotherapyonCBNKcells...
