The multifaceted nature of NLRP12
[摘要] NLRsareaclassofcytoplasmicPRRswithvariousfunctions,rangingfrompathogen/damagesensingtothemodulationofinflammatorysignalingandtranscriptionalcontrolofMHCandrelatedgenes.Inaddition,someNLRshavebeenimplicatedinpreimplantationandprenataldevelopment.NLRP12(alsoknownasRNO,PYPAF7,andMonarch‐1),amemberofthefamilycontaininganN‐terminalPYD,aNBD,andaC‐terminalLRRregion,isoneofthefirstdescribedNLRproteinswhoseroleremainscontroversial.TheinteresttowardNLRP12hasbeenboostedbyitsrecentinvolvementincoloncancer,aswellasintheprotectionagainstsomesevereinfections,suchasthatinducedbyYersiniapestis,thecausativeagentofplague.AsNLRP12ismainlyexpressedbytheimmunecells,anditsexpressionisdown‐regulatedinresponsetopathogenproductsandinflammatorycytokines,ithasbeenpredictedtoplayaroleasanegativeregulatoroftheinflammatoryresponse.Herein,wepresentanoverviewoftheNLRfamilyandsummarizerecentinsightsonNLRP12addressingitscontributiontoinflammatorysignaling,hostdefense,andcarcinogenesis...
