HIV‐1 envelope–receptor interactions required for macrophage infection and implications for current HIV‐1 cure strategies
[摘要] MyeloidcellsresidingintheCNSandlymphoidtissuesaretargetsforproductiveHIV‐1replication,andtheirinfectioncontributestothepathologicalmanifestationsofHIV‐1infection.TheEnvscanadoptalteredconfigurationstoovercomeentryrestrictionsinmacrophagesviaamoreefficientand/oralteredmechanismofengagementwithcellularreceptors.ThisreviewhighlightsevidencesupportinganimportantroleformacrophagesinHIV‐1pathogenesisandpersistence,whichneedtobeconsideredforstrategiesaimedatachievingafunctionalorsterilizingcure.WealsohighlightthatthemolecularmechanismsunderlyingHIV‐1tropismformacrophagesarecomplex,involvingenhancedand/oralteredinteractionswithCD4,CCR5,and/orCXCR4,andthatthenatureoftheseinteractionsmaydependontheanatomicallocationofthevirus...
