Sexual dimorphisms in leukocyte trafficking in a mouse peritonitis model
[摘要] Sexualdimorphismsexistintheincidenceandseverityofmanydiseases,withfemalesdemonstratingrelativeprotectionfrominflammatoryconditions.Theextentandmechanismsbywhichexcessiveleukocyterecruitmentunderliesthesedifferencesarenotwellestablished,andbetterunderstandingisessentialforthedevelopmentoftargetedtherapies.Here,wesetouttocomparethemaleandfemaleinflammatoryresponseinamurinezymosan‐inducedperitonitismodelandtounderstandhowleukocytesubsetsaremobilizedfromstoragepoolsinbothsexes.ThisworkshowsthatfemaleC57BL/6micerecruitfewerclassicalmonocytesandneutrophilsduringzymosan‐inducedperitonitis.Inaddition,sexdifferenceswereevidentinthecirculation,asfemalemiceshowedreducedneutrophiliaandmonocytosisvs.malecounterparts,despitehavingsimilarmobilizationfromBMstores.Importantly,weshowthatstorageandtraffickingofsplenicleukocytesduringacuteinflammationaredistinctbetweenthesexes.Malemicehavegreatersplenicstoresofneutrophilsandclassicalandnonclassicalmonocytes,despitesimilarspleensizes,signifyinganothersourceofpotentialpathogenicleukocytes.Thisworkdemonstratesthatmalesandfemaleshavedistinctleukocyte‐traffickingprofilesinacuteinflammationandsuggeststhatthespleen,nottheBM,playsaroleindeterminingsexdifferencesintheavailablepoolofimmunecells.Suchdimorphismsdemonstratetheimportanceofconsideringgenderinassaydevelopment,drugdesign,andclinicaltrials...
