Lipid raft‐associated β‐adducin is required for PSGL‐1‐mediated neutrophil rolling on P‐selectin
[摘要] Lipidrafts,aliquid‐orderedplasmamembranemicrodomain,arerelatedtocell‐surfacereceptorfunction.PSGL‐1,amajorsurfacereceptorproteinforleukocyte,alsoactsasasignalingreceptorinleukocyterolling.ToinvestigatetheroleoflipidraftinPSGL‐1signalinginhumanneutrophils,wequantitativelyanalyzedlipidraftproteomeofhumanpromyelocyticleukemiacelllineHL‐60cellsandidentifiedalipidraft‐associatedproteinβ‐adducin.PSGL‐1ligationinduceddissociationoftheraft‐associatedproteinβ‐adducinfromlipidraftsandactin,aswellasphosphorylationofβ‐adducin,indicatingatransientuncouplingoflipidraftsfromtheactincytoskeleton.Knockdownofβ‐adducingreatlyattenuatedHL‐60cellsrollingonP‐selectin.WealsoshowedthatSrckinaseiscrucialforPSGL‐1ligation‐inducedβ‐adducinphosphorylationandrelocation.Takentogether,theseresultsshowthatβ‐adducinisapivotallipidraft‐associatedproteininPSGL‐1‐mediatedneutrophilrollingonP‐selectin...
