Perforin‐2/Mpeg1 and other pore‐forming proteins throughout evolution
[摘要] Developmentoftheancientinnateimmunesystemrequirednotonlyamechanismtorecognizeforeignorganismsfromselfbutalsotodestroythem.Pore‐formingproteinscontainingthemembraneattackcomplexPerforindomainwereoneofthefirsttriumphsofaninnateimmunesystemneedingtoeliminatemicrobesandvirallyinfectedcells.MembraneattackcomplexofcomplementandPerforindomainproteinsisuniquefromotherimmuneeffectormoleculesinthatthemechanismofattackisstrictlyphysicalandunspecific.Thelargewater‐filledholescreatedbymembraneattackcomplexofcomplementandPerforindomainporeformationallowaccessforadditionaleffectorstocompletethedestructionoftheforeignorganismviachemicalorenzymaticattack.Perforin‐2/macrophage‐expressedprotein1isoneoftheoldestmembraneattackcomplexesofcomplementandPerforindomainproteininvolvedinimmunedefense,anditisstillfunctionaltodayinvertebrates.Here,wetracetheimpactofPerforin‐2/macrophage‐expressedprotein1fromtheearliestmulticellularorganismstomodernvertebrates,aswellasreviewthedevelopmentofothermembraneattackcomplexesofcomplementandPerforindomainmemberproteins...
