Purinergic signaling during macrophage differentiation results in M2 alternative activated macrophages
[摘要] Macrophagesrepresentahighlyheterogeniccellpopulationoftheinnateimmunesystem,withimportantrolesintheinitiationandresolutionoftheinflammatoryresponse.PurinergicsignalingregulatesbothM1andM2macrophagefunctionatdifferentlevelsbycontrollingthesecretionofcytokines,phagocytosis,andtheproductionofreactiveoxygenspecies.WefoundthatextracellularnucleotidesarrestmacrophagedifferentiationfrombonemarrowprecursorsviaadenosineandP2receptors.ThisresultsinamaturemacrophagewithincreasedexpressionofM2,butnotM1,genes.SimilartoadenosineandATP,macrophagegrowtharrestedwithLPStreatmentresultedinanincreaseoftheM2‐relatedmarkerYm1.RecombinantYm1wasabletoaffectmacrophageproliferationandcould,potentially,beinvolvedinthearrestofmacrophagegrowthduringhematopoiesis...
