TRIF‐dependent TLR signaling, its functions in host defense and inflammation, and its potential as a therapeutic target
[摘要] Toll/IL‐1Rdomain‐containingadaptor‐inducingIFN‐β(TRIF)‐dependentsignalingisrequiredforTLR‐mediatedproductionoftype‐IIFNandseveralotherproinflammatorymediators.VariouspathogenstargetthesignalingmoleculesandtranscriptionalregulatorsactingintheTRIFpathway,thusdemonstratingtheimportanceofthispathwayinhostdefense.Indeed,theTRIFpathwaycontributestocontrolofbothviralandbacterialpathogensthroughpromotionofinflammatorymediatorsandactivationofantimicrobialresponses.TRIFsignalingalsohasbothprotectiveandpathologicrolesinseveralchronicinflammatorydiseaseconditions,aswellasanessentialfunctioninwound‐repairprocesses.Here,wereviewourcurrentunderstandingoftheregulatorymechanismsthatcontrolTRIF‐dependentTLRsignaling,theroleoftheTRIFpathwayindifferentinfectiousandnoninfectiouspathologicstates,andthepotentialformanipulatingTRIF‐dependentTLRsignalingfortherapeuticbenefit...
