Nicotine induces neutrophil extracellular traps

[摘要] NETsservetoensnareandkillmicrobialpathogens.However,NETscanatthesametimecontributetotissuedamageandexcessiveinflammation.Nicotineisamajortoxicagentandhasbeenassociatedwithexacerbatedinflammatorydiseases.Thecurrentstudyaimedatinvestigatingtheroleofnicotine,theaddictivecomponentoftobaccoandelectroniccigarettes,ontriggeringNETformation.WereportthatnicotineinducesneutrophilstoreleaseNETsinadose‐dependentmanner.Nicotine‐inducedNETformationismediatedvianicotineacetylcholinereceptors,dependsonAktandPAD4activation,butisNox2‐independent,asdemonstratedbypharmacologicalinhibitionofNox2andbyuseofNox2‐deficientmouseneutrophils.ThesefindingsdemonstratethatnicotineinducesNETs,whichmayinturncontributetosmoking‐relateddiseases...
